چکیده

BACKGROUND Theophylline was shown to induce contracture development in porcine malignant hyperthermia (MH) susceptible (MHS) skeletal muscles in vitro. The purpose of the current study was to investigate the in vivo effects of theophylline in MHS and MH normal (MHN) swine. METHODS MH-trigger-free general anesthesia was performed in MHS and MHN swine. Theophylline was administered intravenously in cumulative doses up to 93.5 mg·kg⁻¹. The clinical occurrence of MH was defined by changes of central-venous pCO₂, central-venous pH, and body core temperature. RESULTS Theophylline induced comparable clinical alterations in the anesthetized MHS and MHN swine, especially in regard to hemodynamic data. No pig developed hypermetabolism and/or MH according to defined criteria. All animals died with tachycardia followed by ventricular fibrillation. CONCLUSIONS The cumulative theophylline doses used in this study were much higher than doses used therapeutically in humans, as demonstrated by measured blood concentrations. Theophylline is thus not a trigger of MH in genetically determined swine.

Hemodynamic and metabolic manifestations of acute endotoxin infusion in pigs with and without the malignant hyperthermia mutation.

BACKGROUND The hypermetabolic state induced by acute endotoxemia and malignant hyperthermia (MH) may be indistinguishable. The aims of this study were (1) to investigate the differences between MH and sepsis, (2) to determine whether acute endotoxemia can trigger MH, and (3) to establish the effects of dantrolene in these two disorders. METHODS Three groups of swine were studied. All pigs wer...

Altered expression of cardiac myosin isozymes associated with the malignant hyperthermia genotype in swine.

BACKGROUND Anesthetic-induced malignant hyperthermia (MH) in humans and pigs is associated with dramatic alterations in cardiac function. However, it remains controversial as to whether MH-associated cardiac symptoms represent a primary difference of myocardium or a secondary alteration consequent to increases in the hyperthermic stress. Here the authors describe changes in myosin isoform expre...

Effect of vitamin C on tracheal responsiveness and pulmonary inflammation in chronic obstructive pulmonary disease model of guinea pig

Introduction: In the present study, the prophylactic effect of vitamin C, as a potent anti-oxidant, on tracheal responsiveness to methacholine and ovalbumin, bronchoalveolar lavage fluid white blood cell (WBC) count and differential count, and also lung pathology in chronic obstructive pulmonary disease (COPD) -guinea pigs (cigarette exposed guinea pigs) were examined. In addition, the relax...

Plasma dopamine-beta-hydroxylase and platelet monoamine oxidase activities in pigs with different susceptibility to the malignant hyperthermia syndrome by halothane.

Plasma DBH and platelet MAO activities were measured by radioenzymatic assay in 10 Large-White and 20 Piétrain pigs 9 to 11 weeks old. Piétrain pigs within the same litter, challenged by halothane, were classified as malignant hyperthermia (MH) susceptible or not according to their reaction. The Large-White pig strain was not susceptible to MH. Plasma DBH activity did not differ according to st...

Erythrocyte osmotic fragility in hyperthermia-susceptible swine.

Anaesthetic-induced malignant hyperpyrexia in humans appears to result from some intrinsic abnormality of muscle (Satnick, 1969; Harrison et aL, 1970; King, Denborough and Zapf, 1972). Evidence points to this defect being in the sarcoplasmic reticulum (Kalow et al., 1970). A certain strain of Landrace pigs manifests a similar condition and has been used as an animal experimental model for the i...
